Histochemical Fiber Types in the Rat Diaphragmatic Muscle


Abstract
This experiment was designed to assess the fiber types and regional composition of muscle fiber types in the costal and rural portions of the rat diaphragmatic muscle stained with succinic dehydrogenase (SDH) and myosinATPase. It was also performed that non-specific esterase(NS-Esterase), adenylate deaminase (ADA), PAS, H&E and trichrome stains for the classification of muscle fiber types could be available.
The results obtained were as follows.
1. Higher enzymatic activities of SDH, myosin-ATPase, NS-Esterase and ADA were noted in the diaphragmatic muscle than those of Tibialis anterior of the rat. PAS positivity was, however, markedly reduced in the diaphragmatic muscle.
2. Prominent muscle fiber type was type II A fiber, and regional variation of muscle fiber types was noted in the costal and rural portions of diaphragm, especially in sections stained for myosin-ATPase.
3. Identification of muscle fiber types by SDH stain was not always correlated with that by myosin-ATPase stain, especially in the distribution of type II A and I fibers.
4. NS-Esterase stain divided the rat diaphragmatic muscle into three different muscle fiber types according to enzymatic activity. However, ADA stained section showed only two different muscle fiber types. Regional variation of the costal and rural portions was also shown in these enzyme stains.
5. Identification of two different muscle fiber types was also available by conventional H&E, trichrome and PAS stains in the rat diaphragmatic muscle.
